ABSTRACT

Traditional herbal medicine offers opportunities to discover novel therapeutics against SARS-CoV-2 mutation. The dried aerial part of mint (Mentha canadensis L.) was chosen for bioactivity-guided extraction. Seven constituents were isolated and characterized by nuclear magnetic resonance (NMR) and mass spectrometry (MS). Syringic acid and methyl rosmarinate were evaluated in drug combination treatment. Ten amide derivatives of methyl rosmarinate were synthesized, and the dodecyl (13) and 3-ethylphenyl (19) derivatives demonstrated significant improvement in the anti-SARS-CoV-2 plaque reduction assay, achieving IC50 of 0.77 and 2.70 µM, respectively, against Omicron BA.1 as compared to methyl rosmarinate's IC50 of 57.0 µM. Spike protein binding and 3CLpro inhibition assays were performed to explore the viral inhibition mechanism. Molecular docking of compounds 13 and 19 to 3CLpro was performed to reveal potential interaction. In summary, natural products with anti-Omicron BA.1 activity were isolated from Mentha canadensis and derivatives of methyl rosmarinate were synthesized, showing 21- to 74-fold improvement in antiviral activity against Omicron BA.1.

ABSTRACT

The different types of cardiovascular diseases, including coronary heart disease, cardiac arrhythmias and heart failure are highly prevalent in the society. Cardiovascular diseases are the leading cause of mortality. Although the influenza is forced out from the mainstream of thinking nowadays because of the ongoing SARS-CoV-2 pandemic, it still has its serious epidemiological significance. The seasonal influenza epidemic often contributes to mortality mainly, but not exclusively among old, multi-morbid patients. There are a vast number of scientific publications and evidence which prove and emphasize the synergic health-destroying and mortality-increasing effect of co-existing cardiovascular disease and influenza. Moreover, the beneficial effect of vaccination against influenza infection and its major role in prevention is also well documented. The SARS-CoV-2 pandemic enforces the importance of influenza vaccination because both viruses can lead to severe or often fatal disease, especially among old and frail patients. In addition, the younger population can be far more vulnerable against the novel coronavirus in the case of a co-existing influenza infection. International guidelines recommend influenza vaccination for patients having heart disease, like for other high-risk populations. Despite the nationally reimbursed, cost-free vaccines, the influenza vaccination rate of the society is still low not just in Hungary but also internationally. The authors review the effect of influenza infection on heart diseases, and draw attention to the role of influenza vaccination in decreasing cardiovascular morbidity and mortality. ABSTRACT

Recently, there has been an increase in feelings of loneliness and mental health conditions among adolescents. Within this population, parenting teens are at an increased risk for these conditions. Outdoor experiences are shown to be an antidote to loneliness and a way to promote social connectedness by amplifying the processes for supporting social relationships. In 2020-2021, we piloted the 8-week Meeting in Nature Together program (MINT) at a charter school for pregnant and parenting teenagers in Colorado, USA. MINT aimed to promote relatedness and nature connection for students ages 14 to 19. MINT included online and in-person group meetings with educational content, creative activities, discussion, park excursions, mindfulness activities, journaling, and nature photography. Here, we ask, can a school-level nature-based social intervention reduce loneliness among pregnant and parenting teens by promoting and sustaining social connections? How acceptable is MINT to participants? Methods included audiovisual recording transcriptions, surveys, and observation field notes. Results suggest that MINT fostered social connections through a tailored nature-based intervention delivered to a typically isolated community in culturally sensitive, developmentally appropriate ways. MINT proved feasible and effective as participants reported high levels of satisfaction and interest in continuing to engage in activities promoted in MINT.
